I want to upgrade my main SSD C: drive on my Windows 10 laptop.
Can I use Duplcati to copy/clone the existing C: drive (all system files) to a new drive and then swap them out?

No, Duplicati is not suitable for this task at all. It’s designed for backing up user data files. I would not attempt to back up programs or the operating system with it.
Personally I use Macrium Reflect for image level backups. I have also used the backup utility built in to Windows 10 to make image level backups in the past, but I believe it’s been deprecated.
